TCT Receives Patent on Therapeutic Monoclonal Antibody Targeting Pancreatic Cancer
Targeted Cancer Therapeutics LLC (TCT) announced that its anti-CEACAM6 product received the U.S. Designation of Patent Number 8,038,996.

SANTA ROSA, CA, January 17, 2012 /24-7PressRelease/ -- Targeted Cancer Therapeutics LLC (TCT) announced that its anti-CEACAM6 product received the U.S. Designation of Patent Number 8,038,996. The patent claims allow for a method of treating various types of cancer using an anti-CEACAM6 monoclonal antibody that induces apoptosis in cancer cells.

"We designed a monoclonal antibody that binds to CEACAM protein over-expressed on >95% of pancreatic cancer patients. CEACAM6 is also over-expressed in colon cancers, as well as breast, lung ovarian cancers and certain childhood leukemias." stated Daruka Mahadevan MD, Ph.D., Chief Scientific Officer of TCT. "It is a highly specific monoclonal antibody designed to disrupt CEACAM6 function and was tailored for a superior safety profile. There is an unmet need for effective treatments of pancreatic cancer, as it is uniformly lethal and new treatment paradigms are urgently needed."

The antibody was designed as a humanized anti-CEACAM6 single chain variable fragment (scFv) based on the murine Mab 13-1. PEGylation of the glycine-serine linker was used to enhance plasma half-life. The scFv bound CEACAM6 with high affinity, exhibited cytotoxic activity and induced dose-dependent PARP-cleavage in KRas mutated and wild type pancreatic cancer cell lines. Murine PDA xenograft models treated with low doses of humanized scFv-PEG alone elicited tumor growth inhibition, which was enhanced in combination with gemcitabine. Immunohistochemistry of harvested tumors showed significant apoptosis, with inhibition of angiogenesis and proliferation.

In 2010, there were over 40,000 pancreatic cancer cases, of which there were over 39,300 deaths, providing a dismal prognosis. This is attributable to a lack of early diagnosis and effective treatments. Most patients present with unresectable and/or metastatic pancreatic ductal adenocarcinoma (PDA), with a median overall survival of 12 and 6 months respectively.
